Police said their enquiries had "determined that the car was registered to an address in the Corban Avenue area of Enniskillen".
They added that when officers arrived at the house in Enniskillen they discovered the body of the woman.
Det Ch Insp Gary Robinson of the Police Service of Northern Ireland (PSNI) said detectives were working to establish the full circumstances surrounding the woman's death and appealed for information.
"We're especially keen to hear from anyone who was travelling on the Cornagrade Road into Corban Avenue between 18:30 GMT and 22:45 GMT on Saturday and noticed anything out of the ordinary, or suspicious, to make contact with us," he said.
Fermanagh and Omagh District Commander, Supt Pete Cunningham said his thoughts "are with the victim's family and friends as they struggle to come to terms with what has happened".
"I want to reassure the local community that detectives are working at pace, and we would also urge members of the public not to speculate," he said.
"As a victim-focused organisation, violence against women and girls is a priority for policing, and I understand that incidents like this will cause significant concern.
"We are doing everything we can to ensure women and girls feel safe in our communities."
He added that there would "be an increased police presence in the area today, and over the coming days".

Sinn Féin assembly member Jemma Dolan said there was a sense of shock across the community and the town following the woman's death.
"As I was driving into Enniskillen, there actually felt like there was a dark cloud over the town," she told BBC News NI.
"Everyone is going on about their daily life, but yet there's this family who are suffering this huge loss and you know it's a loss that will just impact the whole area."
