Nottinghamshire Police said an investigation was launched following reports a room within the address in Woodborough Road was being used for sexual purposes.
So far, a number of victims have been identified as children, and officers are working to determine the full scale of activity there, police added.
The men, aged 52 and 33, were detained on suspicion of a number of offences, including facilitating the sexual exploitation of children aged 13 to 17.
Insp Vincent Clark, who is leading the investigation, said: "As you would expect, we are taking these reports extremely seriously as we start to unravel what has been happening within this room.
"We are also safeguarding any victims that have come forward who are being supported by our dedicated team of officers.
"It is important to stress that while this property is multi-occupancy, we are not currently investigating any of the people who live at the property.
"The two people we have arrested do not reside at the address. It has been reported that the sexual activity occurred within a rented space in the building.
"We know that this is likely to cause alarm within our local community and officers will be out and about to offer reassurance or address any concerns you may have."
