Truro City will now have to focus on winning the Western League after losing 3-2 at Brislington in the Les Phillips Cup on Saturday. The Cornish side were 3-0 down at half time but rallied late on with penalties from Stuart Yetton and Kevin Wills.
"Full credit to the lads, they never threw the towel in and we really pushed them all the way," assistant manager Glyn Hooper told BBC Radio Cornwall.
"It's really disappointing to go out of another cup competition," he added.
Truro were knocked out of the FA Vase earlier this month by Whitley Bay and now have just the league to concentrate on.
They are currently five points clear of second-placed Corsham Town with a game in hand.
