Chambers will remain at Colchester until the end of the current season.
Colchester United have extended Leo Chambers' loan deal from West Ham to the end of the 2015-16 season.
The 20-year-old defender has made five appearances in League One since joining Colchester in February.
Chambers has appeared three times in the League Cup for West Ham but is yet to make a Premier League appearance for the London club.
He is expected to miss Colchester's game at Coventry on Tuesday because of injury.
