Trinder scored tries in the 10th and 14th minute
Henry Trinder scored two tries as Gloucester ran riot in Rovigo.
Freddie Burns' penalty was followed by two quick tries for Trinder, while Andy Hazell and Charlie Sharples also went over before half-time.
Sharples scored his second try a minute after the break, before Rovigo's first points came from a Younes Anouer try.
Gloucester scored three tries in the final seven minutes - two in three minutes from Tim Taylor, and Jonny May on the stroke of full time.
Gloucester boss Bryan Redpath told BBC Radio Gloucestershire: "With the results last night (Friday) the game was insignificant but I'm delighted the way the boys stuck at it and got some good tries.
"We knew it would be tough and not as easy as it was at Kingsholm. But in general we acquitted ourselves well.
"We scored a few tries and it was a good atmosphere, with more Gloucester fans than Rovigo. Peter Buxton led well from the front, Freddie Burns played very well in terms of his understanding. In general I'm delighted with everyone
"We need to redeem ourselves. I'm bitterly disappointed because I think we could have got through to the quarter-finals and we let ourselves down in that side."
