Southampton teenager Gareth Bale has been added to the Wales squad for Sunday's friendly with a Basque XI. The 16-year-old left-back impressed boss John Toshack on his Wales Under-21 debut against Cyprus on Tuesday.
Cardiff's Jason Koumas has pulled out for "personal reasons", while club-mate Paul Parry has also withdrawn because of a knee problem.
Swansea City midfielder Owain Tudur Jones and left-back Sam Ricketts are unavailable because of club duty.
Uncapped midfielder Jones and versatile full-back Ricketts will be preparing for Swansea's League One play-off final with Barnsley on 27 May.
Toshack - who has strong Swansea ties - had always hoped both would be unavailable for the game, but he is unlikely to be impressed by the latest of a series of late withdrawals from Koumas.
The Basques have called Bolton defender Ivan Campo into their squad for Sunday's match, who is the only foreign-based player.
Middlesbrough's Gaizka Mendieta is still injured, while Liverpool's Xabi Alonso and Asier Del Horno of Chelsea are with the Spain national team preparing for the World Cup.
However, Alonso's brother, Mikel, the Real Sociedad midfielder is included.
There is no place for Everton's Mikel Arteta in a squad drawn from the ranks of Sociedad - Toshack's former side - Athletic Bilbao, Osasuna and Alaves.
Bale, a former room-mate of England's World Cup-bound sensation Theo Walcott, became Southampton's second youngest player when he made his debut in the Coca-Cola Championship last month.
And he showed enough promise on his Wales U21 debut as a left-sided midfielder to convince the on-looking Toshack that he has the ability to handle top-class international football.
MK Dons defender Craig Morgan and Verona striker Craig Davies have also been promoted from the U21 squad.
Bale, Morgan and Davies will return to the U21 squad for next Wednesday's second leg Uefa Championship qualifier with Estonia at Wrexham.
They will joined by fellow U21 stars Lewis Price and Andrew Crofts, but Lewin Nyatanga, Joe Ledley and David Cotterill will stay with the senior squad for three days training in Spain after the non-capped Basque game.
The squad will then fly to Austria on Thursday to prepare for Saturday's friendly with Trinidad and Tobago in Graz.
