Earlier on, Steve Croft fired 65 and Tom Smith 44 to lead the visitors to 206-8 and a lead of 301 runs, setting up a declaration before tea.
And Muralitharan had Sussex in all sorts of trouble with a superb spell.
But stubborn knocks from Chris Nash (35) and Richard Montgomerie (50) proved vital, taking Sussex to 145-5.
Set 302 to win in a minimum 66 overs, Sussex opted not to chase the total and were content to shut up shop as best they could on a pitch of good pace and bounce.
Montgomerie and Nash shared a stand of 93 in 43 overs, before Muralitharan struck twice in two balls to spark the loss of four wickets for 15 runs in four overs.
But it came too late for the Sri Lankan off-spinner, who had to wait 13 overs to take his first wicket.
