England's winter sensation James Anderson took the first five Surrey wickets as Lancashire became the first county since 1998 to ask the champions to follow on.
Anderson took 5-61 and leg-spinner Chris Schofield mopped up the tail with three wickets as Surrey had to bat again 319 runs behind.
Ian Ward (49), just before lunch, plus Mark Ramprakash (50) and Graham Thorpe (15) all suffered lapses of concentration to gift Anderson his second, third and fifth wickets.
But his first success, a vicious swinging yorker which uprooted Jon Batty's off stump, and the ball which nipped back to trap Ali Brown lbw, were both superb pieces of bowling.
Following on, Batty again fell cheaply, this time to Glenn Chapple, and Andrew Flintoff dismissed Ramprakash before rain and bad light checked Lancashire's progress.
Surrey may need the weather to come to their assistance on Monday, or else Ward and Thorpe will have to bat with plenty of nous to avoid defeat.
