Matthew Hayden scored 177 and Justin Langer 111 as Australia set the West Indies a daunting target of 418 to win the fourth and final Test.
The home side made a solid start to their run chase to reach 47-0 at the close after finally bowling out Australia for 417.
But the damage had already been done by Hayden and Langer's first-wicket partnership of 242.
The West Indies will now have to score their highest-ever fourth-innings total against Australia to stop the tourists becoming the first side to achieve a clean sweep in the Caribbean.
Hayden survived two let-offs to reach his 14th Test ton, and Langer brought up his 15th century soon after.
Langer then edged Chris Gayle to Brian Lara at first slip and Adam Gilchrist made just six before he was caught by Marlon Samuels off Omari Banks.
Onslaught
Banks and Samuels again combined to remove Martin Love for two on the last ball of the morning session.
Hayden continued his onslaught after lunch, hitting a huge six off Banks.
Mervyn Dillon then bowled Darren Lehmann, who got an inside edge on to his stumps for 14.
And Hayden was run out by Carlton Baugh after Waugh played a shot to backward point and sent him back.
Dillon completed a four-wicket haul by dismissing Andy Bichel for a duck, Brett Lee for 18 and Stuart MacGill without scoring.
Vasbert Drakes then picked up the wickets of Jason Gillespie (5) and Glenn McGrath (14), leaving captain Steve Waugh on 45 not out.
West Indies openers Devon Smith and Chris Gayle were unbeaten on 21 and 19 respectively when bad light forced play to be stopped.
