They started well but lacked the killer instinct of their opponents who looked dangerous with the ball in hand.
Spencer missed an early penalty but made amends soon after, but that was the last time Gloucester led.
Waldouck cut through the Gloucester midfielder with worrying ease and crossed for his side's first try.
Dave Walder converted with Spencer sending over a penalty soon after.
But two penalties from Walder gave the visitors a 13-6 lead at the break.
Things got no better for Gloucester after half-time with a flowing three-quarter move seeing Van Gisbergen go over in the corner after the Wasps forwards had driven their side into Gloucester's 22.
Walder missed the conversion but later sent over a penalty before Joe Simpson bumped off Gloucester replacement Nicky Robinson to pass to Betsen who went over under the posts unopposed.
Again Walder converted as he did for Van Gisbergen's late try in the corner as Wasps cemented an impressive result.
The only negative for Wasps was an injury to Waldouck who suffered a suspected fractured cheekbone after a clash of heads midway through the second half.
Gloucester coach Bryan Redpath:
"We didn't create enough pressure to get points. We won't hide from the fact that it was a poor performance and, if you perform like that, you are never going to compete.
"They were excellent and defended well. We are lacking massively in confidence and I don't question our work ethic but there are a lot of things to do.
"Everyone has to be realistic and know what their part in the game is. If someone has not got a clear understanding of it, they must say. Sometimes people hide behind barriers.
"For me, it is a tough time in my career as a coach. I won't hide from my responsibility and I certainly won't hide from taking responsibility for this heavy defeat at home which is disappointing."
Wasps director of rugby Tony Hank: "The victory was built on the way we defended in the first half and the way we responded to the pressure.
"Our effort and intensity in those 50-50 situations was good."
He added: "Dominic has gone to hospital to look at his cheekbone. That would be a big loss to us if we lost him."
