Wigan gave their fans an end-of-season send-off at the JJB Stadium to raise questions about Hull's play-off hopes.
Iafeta Paleassina stretched over for the opening try and the superb Michael Dobson, and Chris Ashton with two, added further first-half tries.
Matt Dale crossed for Hull but further tries from Harrison Hansen and Nathan McAvoy put the outcome beyond doubt.
Hull did improve in the final quarter and restored some pride with scores from Motu Tony and Shaun Briscoe.
But Gareth Hock had the last word with Wigan's seventh try late on, concluding their eighth successive home win.
Wigan coach Brian Noble:
"It's certainly been an interesting season. "We were the team that was bottom at the halfway point in the season - and we didn't finish bottom.
"The way we finished is testament to how much hard work we've done and I can't thank the players enough."
