New Orleans star Reggie Bush will miss the clash with San Diego at London's Wembley Stadium next Sunday after injuring his left knee.
Running back Bush will have surgery on Monday after missing the second half of Sunday's 30-7 loss to Carolina.
"We're going to miss him, and I hope he's not out very long," said Saints quarterback Drew Brees.
Chargers back LaDainian Tomlinson again struggled with a toe injury, carrying for only 41 yards in a loss to Buffalo.
The game at Wembley was expected to feature two of the league's most exciting stars in Bush and Tomlinson, and two of its leading teams.
But both sides have won three games and lost four so far this year. The Saints sit at the bottom of the NFC South division, while San Diego are second in the weak AFC West.
Both teams were due to arrive at Gatwick Airport early on Monday, with New Orleans set to use Arsenal's training ground this week while San Diego head to Pennyhill Park in Surrey, a facility the England rugby team often use.
The game will be screened live on BBC TV and - for UK users - on the BBC Sport website, with former NFL star Jerry Rice the studio guest.
Wide receiver Rice won three Super Bowl rings with the San Francisco 49ers in a career that lasted a remarkable 19 years.
