Omagh boss Roy McCreadie had goaded the Glens with a pre-match prediction that they might blow their lead but was forced to eat humble pie afterwards.
Andy Smith scored the first two and Darren Lockhart the third as Glentoran stormed into a 3-0 half-time lead.
Colin Nixon headed number four as the Belfast side maintained their nine-point lead at the top of the Premier Division.
McCreadie's team selection again raised a few eyebrows.
Eamon Kavanagh, James Quigley, Marty McCann and Ivan Sproule, who all featured in Thursday's Irish Cup win over Linfield, did not make the starting line-up.
Smith's first goal came after just three minutes when he flicked on Gary Smyth's free-kick.
The second was in the 34th minute when Scott Young's left-wing cross was superly headed in by Smith.
Darren Lockhart pounced from close-range for Glentoran's third a minute later.
Nixon's back post header on the hour put the icing on the cake for a rampant Glentoran side.
