Pat Fenlon celebrated a superb win in his first match as Derry City manager as the Eircom League side scored the only goal late on at the Oval. Kevin McHugh got the vital strike in the 85th minute, running down the right before firing the winner past Glen goalkeeper Elliott Morris.
Glentoran, whose only win the 2006 competition was at home to Derry, had a string of chances in the first half.
But Michael Halliday, Dean Fitzgerald and Paul Leeman could not find the net.
Gary Hamilton also shot inches wide while Tim McCann sent a 30-yard effort just over.
For City, Pat McCourt shot just too high and McHugh forced two fine saves from Morris.
City boss Fenlon was ordered from the touchline, along with Glentoran assistant boss Andy Mathieson, after becoming involved in a touchline dispute.
"I was angry about a tackle on Pat McCourt and was surprised the referee saw fit to send me off," said Fenlon.
"I was not sent-off at all last season and feel I have let the players down."
