Dartford, Dover Athletic and Bromley are all seeking a third straight win in Blue Square Bet South on Saturday.
Bromley host Maidenhead, Dover make the 650-mile round trip to Truro City while Dartford host Hampton & Richmond.
Darts manager Tony Burman told BBC Radio Kent: "It's a tough two weeks, I said 10 points from the first six. Hopefully we come through again."
Elsewhere Hythe Town play their first ever Ryman League game against Godalming Town.
Hythe won the Kent League title last season in the same season they reached the first round proper of the FA Cup so are used to playing with bigger sides.
I don't want to see the players sitting around a hotel on Friday night and Saturday morning being bored so there won't be any overnight stays for us this year
Dover boss Martin Hayes
Manager Scott Porter said: "It's a massive achievement for myself, the club, and the players who got us there. When you get there you think 'Wow, this is a big step up' but there's been a lot of hard work on and off the pitch and we have now got our rewards by being in the Ryman League."
Dover Athletic have decided not to take an overnight stay to their long trip to Cornwall, but will take a return flight from Manston Airport on the day as both they and opponents Truro seek their third straight win.
Dover boss Martin Hayes said: "The chairman looked at all the possibilities and decided to go down on Saturday. There are clubs who will do an overnight stay but I don't want to see the players sitting around a hotel on Friday night and Saturday morning being bored so there won't be any overnight stays for us this year.
"It's a hell of a trip, but a lot of the lads who played in the Conference are used to long trips. They've got a lot of experienced players, Barry Hayles up front may not have the legs anymore but he knows what to do and when to do it."
Hayes refuses to get carried away by winning both their first two games 4-0, adding: "It's been a good start in terms of results, so I can't complain, but we know we haven't played brilliantly. We've made mistakes and not been punished but I'll take two 4-0 wins as a base."
Kent enters the FA Cup on Saturday with 13 teams starting the extra preliminary round, needing six wins to reach the first round proper.
Also a new football league starts on Friday night as the Kent Invicta League, in step six of the football pyramid, begins with the inaugural game, Erith and Dartford Town against Phoenix Sports. The pick of the other games in that division sees Ashford United, newly reformed after the demise of Ashford Town, visit Kent County League champions Hollands and Blair.
