Kyle Perry put The Lambs ahead with a header from Scott Barrow's cross and the lead was doubled when Tom Marshall netted from close range.
Danny Thomas' shot crept over the line for 3-0, before Eastbourne pulled one back with Richard Pacquette's header.
Alex Rodman tapped in for 4-1 as Tamworth claimed the biggest win of the day in the division, despite a late consolation header from Gary Elphick.
