James Constable scored twice and made one as Oxford overcame Rushden to move to within two points of the play-offs.
Oxford went ahead with the first chance when Craig Nelthorpe crossed for Constable to finish from 12 yards.
An improved Diamonds equalised on the hour, Robert Wolleaston lobbing over Billy Turley from Lee Tomlin's pass.
Constable restored the lead with a penalty after he was brought down by Chris Hope and then put in Simon Clist to seal the win in the dying seconds.
