Carlisle destroyed Macclesfield's resistance with a second-half burst to secure their place in the quarter-finals of the Johnstone's Paint Trophy.
Shaun Brisley headed Town in front, but Matty Robson equalised from the angle and Scott Dobie flicked in for 2-1.
Marc Bridge-Wilkinson then added a third with a downward header and Dobie grabbed his fourth in four games in the 80th minute with a neat 12-yard finish.
John Rooney curled in a consolation, but it was Carlisle who progressed.
