Mitchell's hundred followed back-to-back centuries against Gloucestershire
Daryl Mitchell struck his third consecutive century to lead a Worcestershire fightback on day two.
His unbeaten 136 underpinned a revival for the visitors who were reduced to 38-3 early on.
Alexei Kervezee (72) and Shakib Al Hasan (47) offered good support as Worcestershire rallied to 281-6.
Earlier Dean Cosker (41 not out) and Huw Waters compiled a half-century stand for Glamorgan's last wicket as they moved to 369.
That left Worcestershire needing 220 to avoid the follow-on and they were quickly in trouble when Robert Croft had David Wheeldon caught by Jamie Dalrymple at slip with his fifth ball of the match.
Cosker then trapped Vikram Solanki lbw with the first delivery of his spell.
The visitors' problems increased with the second ball after lunch as Moeen Ali fell lbw to David Harrison.
But the arrival of Kervezee halted Glamorgan's momentum as he and Mitchell set about constructing a fourth-wicket partnership of 145.
Kervezee was eventually caught at extra cover by Tom Maynard off Jim Allenby.
But Al Hasan then combined with Mitchell for a further 92 runs before he fell to another Maynard catch off the bowling of Waters.
