Chapple finished the day with two wickets for 23 runs
Glen Chapple and James Anderson gave Lancashire control over Durham, despite rain permitting only 21 overs on day three at Chester-le-Street.
Chapple removed Paul Collingwood and Ian Blackwell, with Anderson accounting for Michael di Venuto, as Durham, who had resumed on 27-1, slipped to 53-4.
Dale Benkenstein (21) put up some resistance, but fell lbw to Tom Smith.
Phil Mustard and Ben Stokes were left to see Durham to the close on 96-5, still 99 shy of avoiding the follow-on.
Collingwood resumed his innings on a murky morning and was immediately under pressure from his England colleague Anderson, making his way to 12 before Lancashire skipper Chapple had him caught at second slip.
Chapple then cleaned up Blackwell just as the drizzle began to fall and play stopped for the first time.
On the resumption, Durham added a further 38 runs as Anderson and Chapple continued to bowl unchanged.
The skipper finally made way for Tom Smith who trapped Benkenstein in his second over.
Stokes and Mustard both looked secure at the crease when the rains returned for good.
BBC Radio Newcastle's Martin Emmerson: "The England players on show here on day three suffered differing fortunes. Paul Collingwood had a torrid time at the hands of his international team-mate Jimmy Anderson.
"Anderson and Lancashire captain Glen Chapple both bowled with real ferocity during the 20 overs of play that was possible as the ball swung around incredibly in the damp conditions.
"After Anderson struck Collingwood on the arm with a short delivery it was Chapple who finished him off. The batsman, bruised and no doubt still wondering about the effects of the blow, edged to Tom Smith in the slips for 12.
"Durham crumbled, losing four wickets for 69 runs as Michael Di Venuto, Ian Blackwell and Dale Benkenstein all fell in a spell of bowling which made the ball unplayable at times.
"The weather eventually intervened in timely fashion, and who knows? It may yet save Durham, but if Lancashire serve up this type of ferocious bowling again on the final day they could still win this and bowl Durham out twice."
