Warwickshire wicket-keeper Tim Ambrose struck his maiden double century to put the Bears in command, Worcestershire closing day two 494 behind at 116-2.
The 24-year-old, born in Australia but now qualified for England, hit 34 fours in an unbeaten 251, the highest in the Championship by a keeper for 10 years.
He shared 226 with Heath Streak before Darren Maddy declared on 610-6.
Dale Steyn then ousted Australia's Phil Jaques for a 10-ball duck, but Stephen Moore lifted the home side with 46.
Maddy's declaration denied Ambrose the opportunity to eclipse Geoff Humpage's Warwickshire wicket-keeping record of 254 against Lancashire at Southport in 1982.
He did share in a record sixth-wicket partnership, beating the 220 scored by Tom Dollery and Jack Buckingham against Derbyshire in 1938.
