Klusener hit 131 off 160 balls to take the visitors to 369 in reply to Glamorgan's 204, before the hosts ended the day on 65-2, still 100 runs behind.
Young seamer Huw Waters had a good day with the ball for Glamorgan, taking 4-33, but other bowlers suffered.
Monty Panesar, 6-80 in the match to date, could force a Saturday victory.
Glamorgan were not helped when slow left-armer Dean Cosker limped off after slipping in the
field, having taken two Northants wickets.
