Glamorgan stalwart Robert Croft captured 13 wickets in the match to inspire his team to a 10-wicket win.
Gloucester began the final day needing 174 to make Glamorgan bat again with eight wickets intact, and Hamish Marshall and Alex Gidman hit fifties.
But Croft took two wickets in his first over to secure Glamorgan's third success in a row at Cheltenham.
The visitors had to make 41 from 15 overs but needed less than four of them to complete an impressive victory.
There were fears of rain in the Cheltenham area, but only two overs were lost to a light shower.
Croft's match return of 13-187 was one wicket short of his personal best against Warwickshire at Swansea 14 years ago.

Marshall's 53 contained six boundaries and spanned two hours, but Croft got one to bounce and it created a catch for leg-slip, while Gidman also fell to the former England spinner having batted for over two hours.
Ryan Watkins equalled a Glamorgan record with five leg-side catches.
Brendon McCullum then crashed two sixes and five fours in 34 from 13 balls as Glamorgan finished the match swiftly.
The Dragons' captain Croft was keen to pay tribute to the efforts of the entire team.
"The responsibility was on our batsmen after winning the toss because you'd expect a pitch like this to turn later in the game," the off-spinner told BBC Sport Wales.
"Mike Powell and David Hemp took real responsibility. We got over 600 and were able to apply pressure from there.
"We kept our patience, and the catching around the bat was fantastic, especially from Alex Wharf and Ryan Watkins.
"Personally, I feel that I'm bowling as well as ever. I'm happy with my action and my body is feeling fine."
