Hampshire took an iron grip over Nottinghamshire side on day two of the Division One match at the Rose Bowl.
The hosts began the day on 345-5 which became 401 all out, Charlie Shreck swinging the ball well to take 5-94.
Then Dimitri Mascarenhas destroyed the Notts top order in figures of 4-25. David Hussey's 26 was the best score in a feeble first innings total of 147.
Hampshire captain Shane Warne did not enforce the follow-on, and his team were 148-1 at stumps, 402 ahead.
John Crawley was 63 not out, making progress towards his second century of the match, with Jimmy Adams 53 not out.
Afterwards, Mascarenhas said he was "a little bit disappointed" not to be named in the squad for England's one-day series against Sri Lanka.
He said: "I've not really had a great season so far so I didn't really warrant a spot."
