Yorkshire resumed on 15-0 but lost both openers to Heath Sreak, before South African-born Tim Groenewald claimed two wickets on his Championship debut.
Having seen Michael Lumb dropped in his first over, he had him caught low at slip, then ousted Darren Lehmann.
Anthony McGrath's 48 took the Tykes to 216, but Nick Knight and Jon Trott put on 85 to increase the home advantage.
Having claimed his first wicket, Groenewald, who qualifies through a British passport, then saw Craig White dropped by wicketkeeper Tim Ambrose with his next delivery.
He ended White's innings later when he deflected the ball onto the stumps in his follow-through to run out the Yorkshire captain at the non-striker's end.
"It's been awesome. I tend to know quite early in my spell if it's going to be a good day or whether I'm going to have to work quite hard, and after my first over I felt it was coming out quite nicely and I knew it was going to be good.
"You just have to make the most of it when it's like that," Groenewald commented.
Neil Carter made a dramatic start to the Bears reply, launching Australian Test bowler Jason Gillespie for two sixes in his second over, before he was caught on the point boundary for 36 from 32 balls.
Meanwhile, England captain Michael Vaughan continued his recovery from injury by practising his throwing in the nets.
