The hosts were finally all out for 534 at Sophia Gardens, with Jamie Dalrymple hitting 108, Peter Trego thrashing 72 and David Harrison taking 5-117.
Glamorgan's first innings relied on some late hitting by Robert Croft (84) who was finally dismissed by Ed Joyce.
Following on, the Welsh county were still way behind on 79-2 at stumps.
