Harbhajan Singh confirmed his return to fitness with four wickets for a Rest of India side in Madras. The spinner, who has been out of action with a finger injury since the World Cup, grabbed 4-79 to bowl domestic champions Mumbai out for 244.
Tendulkar himself made 50, taking his match aggregate to 144 in his first-class return from finger surgery.
Ramesh Powar underlined his all-rounder credentials, top-scoring with 57, as the Rest were set 340 for victory.
Test opener Sanjay Bangar fell in the second over and and Virender Sehwag followed for 36, casting further doubt on the future of their partnership.
But Rahul Dravid provided a solid 16 from 49 balls to see his side to stumps at 55-2.
The Irani Trophy is the traditional season-opener, and provides a Test trial ahead of the series against New Zealand.
