Two first-half tries by Danny McGuire helped Leeds to second place in the table but only after they endured a strong fightback by Harlequins.
McGuire's brace, either side of a score by Brent Webb, put the Rhinos 20-4 ahead at the break as Quins paid the price for a sluggish start.
Chris Melling's try was Quins' lone first-half score, Matt Gafa adding another after Webb scored his second.
Kevin Sinfield had kicked Leeds to safety before Paul Sykes's late try.
Leeds coach Tony Smith on Brent Webb: "Webb is just a world-class player. He's just unbelievable with some of the things he does and he's got the ability to put balls on a plate.
"He's been something special and he's one of the better players that we have seen come over here and perform.
"Harlequins are always like that and we knew we were in for a tough match."
Harlequins coach Brian McDermott on his side's poor start:
"It put us 12 points down and that was always going to make it difficult for us.
"I thought we did enough to win the match, although Leeds were outstanding near their own try-line.
"There's a lot of character in the side and the boys are sat in there devastated because it's our fourth straight loss."
