Durham closed day three on 10-0, chasing a daunting target of 361 to beat Derbyshire at the County Ground.
Tom Lace scored a career-best 61 and Matt Critchley made 51 as Derbyshire built an imposing lead.
Harvey Hosein (62) also chipped in with a half-century - his second of the match - before the hosts were bowled out for 334 in their second innings.
Durham openers Tom Lees and Gareth Harte survived the final 14.2 overs of the day before bad light stopped play.
Derbyshire's Tom Lace, who scored his first half-century in first-class cricket, speaking to BBC Radio Derby:
"It's a monkey off my back. There were a few times last year I got close. I had a tough pre-season, so it's nice.
"It worked out pretty well, because when Harvey [Hosein] first came in, he upped the tempo and we had a nice flurry of boundaries before tea, and then after tea, Harvey did the role of occupying the crease and Matthew Critchley took that [attacking] role. I thought they complimented each other brilliantly.
"If you'd said to us after the first five minutes of play on the first day that we'd be in this position, we'd have bitten your hand off."
