Sheffield United lost at home for the second time in four days as Colchester held off a second-half fightback.
George Moncur gave the U's a 2-0 lead at the break with two smart finishes.
The Blades pulled one back when Billy Sharp scored from the spot after Gavin Massey tripped Jay McEveley in the area before Martyn Woolford levelled.
Sordell made it 3-2 before George Elokobi saw red for fouling Sharp in the area but Jamie Jones kept out his penalty.
