Former Olympic 10,000m bronze medallist Brendan Foster founded the event.
Many celebrities will be taking part in the run, including Carol Vorderman who will be "Walking for Whiteley" in memory of Richard Whiteley.
Rail operator GNER will set on extra trains on the East Coast mainline for people travelling to the event.
Closures and parking restrictions will also affect many roads on Tyneside during the event.
