Bath flanker Scott Gray will earn his first cap for Scotland against the Barbarians on Saturday.
The 26-year-old has previously played for Zimbabwe Under-21s but qualifies for Scotland through his father.
Scotland coach Matt Williams could also blood Stephen Cranston and Gareth Morton of The Borders after both were named on the bench.
"Openside is the position that we are looking at and I am excited about specialising in that role," said Gray.
The backs are unchanged from the team that lost in the last Six Nations game against Ireland in March.
But, in the pack, Sale Sharks' Iain Fullarton replaces Stuart Grimes at lock and Cameron Mather and Gray start in the back-row.
Scotland coach Matt Williams admitted he was humbled by the prospect of taking on the Baa Baas.
"The Barbarians are what I consider a magnificent team and it is an honour to coach against them, and to coach against Bob Dwyer. He is a mentor of mine," said Williams.
"I certainly would not be coaching at the top level without Bob and without his support as I was coming through as a coach.
"I know they will come out to play in the greatest tradition of Barbarian rugby so it will be a fantastic afternoon."
Meanwhile, Ross Beattie has been forced to withdraw from the summer tour party with a thigh injury.
His place will be taken by Edinburgh prop Craig Smith for the tests against Australia and Samoa.
