Nicholas Thompson took advantage of heavy overnight rain to shoot an eight under par 64 and take the lead at the 84 Lumber Classic in Pennsylvania.
But Michelle Wie struggled against the men again, shooting a 77.
Thompson, 181st on the US tour money list, recorded tournament's lowest round since Vijay Singh's opening-round 64 in 2004 to claim a two-stroke lead.
He said: "Had the scores been this low without rain I would've been surprised, but the rain softened up the greens."
While there were numerous scores in the 60s, Wie appears set to miss the cut in a US PGA Tour event for a sixth straight time.
Wie's last six rounds against the men include two 79s, a 78 and two 77s.
Thompson, a former US Walker Cup team member, has a two-shot lead over a group of six at 66 that includes Rory Sabbatini and champion Will MacKenzie.
