The lofty Swede romped into the Ryder Cup reckoning with two victories in Europe this season. The 37-year-old makes his first appearance after being controversially left out of the 1999 side.
He was 11th in the rankings, one position away from an automatic spot, but captain Mark James instead handed Scot Andrew Coltart a wildcard.
Karlsson, who is one of the tallest players on Tour at 6ft 5in, also won five European titles before this year.
Karlsson, who has played on two Seve Trophy teams, clinched the 116th and final European Tour card at the end of 2004, pipping Australian Jarrod Moseley by �9.39.
