Southend hit back from a goal behind to leap-frog 10-man Oxford United in the League Two table.
James Constable put the visitors ahead when he converted Jake Wright's well-worked free kick.
The hosts hit back when Ryan Hall controlled and hit a half-volley into the net after an Anthony Tonkin error.
Bilel Mohsni won it when he poked home from close range, as Oxford's misery was compounded when Damien Batt saw a straight red for fouling Anthony Grant.
Southend United manager Paul Sturrock told BBC Essex: "I think the first 15 minutes, we started a bit sluggishly, and also the system we started with didn't suit playing against their system.
"As soon as we changed to 4-4-2 we looked a much more balanced side against their system and I think we dictated the game from then on.
"A lot of things haven't gone our way here this season, so it's nice to think we're getting a wee bit of luck and the rub of the green.
"But you get luck by work rate, attitude and doing the right things."
