Ryan Clarke saved a late Cheltenham penalty to earn Oxford a point.
The Robins took the lead when Wes Thomas chipped the ball over Clarke but Tom Craddock headed in Damian Batt's cross to draw the U's level.
Both sides had goals ruled out, Oxford for a push on keeper Scott Brown and Cheltenham for offside.
And the visitors had the chance to steal the win when Anthony Tonkin fouled Brain Smikle but Clarke's superb dive kept out Thomas's spot-kick.
Cheltenham Town boss Mark Yates told BBC Radio Gloucestershire: "I thought the lads were superb today against a really good Oxford side. We had to defend for our lives in the second-half, but the main thing is that we know we can do it.
"There's a sense that we got something back after a disappointing performance on Tuesday.
"We defended heroically and looked lively on the break and I think we've shown today that we can play and compete which has got to continue."
