Port Vale scored twice in the opening six minutes to notch their first win since the departure of Micky Adams.
John McCombe thumped home the opener from close range before Justin Richards headed home a Louis Dodds cross.
Substitute Shaun Harrad reduced the arrears shortly after the break after he met Jacques Maghoma's low cross.
But Vale held on despite being reduced to 10 men for the second successive match, captain Marc Richards shown a straight red for violent conduct.
Port Vale caretaker manager Geoff Horsfield told BBC Radio Stoke: "They looked like they all wanted to wear a Port Vale shirt whereas on Saturday I don't think some of them did.
"We said that from the first minute we had to hustle them and stop them from playing because we know Burton are a very solid outfit.
"It's been a traumatic week for the players, I don't know if it effected them on Saturday or not because that wasn't them on Saturday".
Burton Albion manager Paul Peschisolido told BBC Radio Derby: "That was the poorest we've ever started.
"We talked about how important it was to come here and make sure we were solid in the first half hour to cause a bit of unrest because there's lots going on at this football club.
"It would have been nice to have been 30 or 40 minutes into this game without having conceded any chances and especially goals, but we shot ourselves in the foot. Seven minutes gone, you're 2-0 down and you're up against it."
