Chesterfield endured a nervy second half to hang for a win which keeps them in the play-off places.
A Robert Page shot struck the crossbar before Aaron Downes slammed in the rebound from close range for the lead.
Dominic Green's deflected 20-yard shot wrong-footed Vale keeper Chris Martin to double the Spireites' advantage.
Craig Davies gave Vale hope after the restart when he struck from 12 yards, however he then wasted four chances to equalise as the visitors held on.
Chesterfield manager John Sheridan told BBC Radio Sheffield:
"They're a good side and they put us under a lot of pressure.
"We got ourselves in a winning position with two goals up and it always gives you a chance.
"We battled hard and defended well. To be honest it's a great result."
