Jacques Maghoma's first-half goal brought Port Vale's six-game unbeaten run to an end at the Pirelli Stadium.
Burton keeper Artur Krysiak blocked a Louis Dodds' effort early on but former Tottenham youngster Maghoma broke the deadlock with a fine shot on the turn.
The home side survived a penalty appeal after Marc Richards' overhead kick appeared to hit the arm of a defender.
Craig Davies almost pulled a goal back midway through the second-half but his angled shot hit the post.
