Chesterfield were left to rue a missed penalty as struggling Lincoln recorded their first win in 10 games.
Cian Hughton, son of Newcastle manager Chris, opened the scores with a far post header from Delroy Facey's cross.
Facey added a second when he looped a header over keeper Mark Crossley and he almost netted again from 25 yards.
Scott Bowden pulled a goal back late-on with a right-footed shot but Jack Lester missed a chance to level when he blazed his 87th-minute spot-kick over.
Lincoln City manager Chris Sutton told BBC Lincolnshire:
"It was important that we got a win under our belts but more importantly I felt we deserved to win, we played some decent stuff and scored two good goals.
"We might have had a stroke of luck at the end with the missed penalty but on balance of play and chances created, we deserved it."
Chesterfield boss John Sheridan told BBC Radio Sheffield:
"I don't think we played that great. You need to win your away games.
"Something needs to be done. Players away from home aren't the same players."
