Jack Lester's ninth goal of the season ended Chesterfield's run of three defeats as Cheltenham's problems go on.
The injury-racked visitors, who parted company with Martin Allen on Friday, rarely threatened and have now won only one of their last 14 matches.
Lester struck when he was found unmarked by a Kieran Djilali corner, scoring with a near post header.
Chesterfield dictated, Cheltenham's only chance coming when Theo Lewis forced Mark Crossley into a save.
Cheltenham acting manager John Schofield told BBC Radio Gloucestershire:
"We battled and scrapped first half and it was ugly, but I asked the lads to stick in there.
"Yes we lost 1-0, but in regards to the test, I think we passed it."
Chesterfield manager John Sheridan:
"I thought we possibly should have scored more goals and made the game a lot easier.
"But, the most important thing was getting the three points after three defeats, and I think we well deserved to win the game."
