Top scorer David Mooney fired Colchester United to within a point of the League One play-off zone with a late winner to see off Peterborough.
Posh were ahead in 35 seconds as Tommy Rowe headed in from close range -the fastest goal in League One this season.
But Steven Gillespie levelled with a low shot into the bottom right corner.
And, with just four minutes left, Mooney struck from six yards with his 10th goal of the season to lift John Ward's U's two places up to ninth.
Posh stay sixth after a second defeat in three games for Darren Ferguson's men since he returned to London Road.
Ferguson's men had made a dream start when Rowe headed in Chris Whelpdale's far-post cross - and the visitors had further chances.
Skipper George Boyd went close before Colchester goalkeeper Ben Williams denied Lee Tomlin from point-blank range, while Whelpdale's header then struck the foot of the post.
But the U's equalised eight minutes before the break, Gillespie converting from close range after Posh defender Kelvin Langmead had only partially cleared Lee Beevers' low cross.
Posh then hit the woodwork for a second time when Charlie Lee's 25-yard effort crashed against the underside of the bar in the opening minute of the second half.
But visiting defender Ryan Bennett had already been forced to clear David Perkins' diving header off the line before Mooney slotted in to end a run of seven games without a goal.
Colchester United manager John Ward told BBC Essex:
"I feel really good about that one. It's a famous win.
"It's one of those wins that you remember for some time, because we've beaten a good team who have played well against us.
"We had a lot of defending to do and there was a major question mark after the first minute, not just for the team but for the supporters.
"But, if you're a Colchester supporter you'll be really pleased."
Peterborough United boss Darren Ferguson said:
"It's a game we thought we should have won.
"I thought we started really well and the game should have been over before they equalised.
"But we conceded two poor goals. We had no composure and both goals were easily avoidable."
