Liam Feeney's volley ensured Lee Bradbury secured his first win as caretaker boss as the Cherries closed in on League One leaders Brighton.
Feeney put the Cherries ahead in the second half, volleying home Rhoys Wiggins' cross from inside the area.
Glenn Murray had the ball in the back of the net for the Seagulls, but his header was ruled offside.
Bradbury's side are now three points off Albion and move above Huddersfield and Southampton into second place.
A sell-out crowd turned up to see the Cherries' first home game since the departure of former boss Eddie Howe to Burnley.
And Feeney ensured Bradbury got a winning start at Dean Court as his thunderbolt produced Bournemouth's first goal under the charge of their veteran utility man.
Having had his header ruled out for offside, Brighton substitute Murray should have put the Seagulls back on level terms, but he could only fire over the crossbar when inside the Cherries area.
Bournemouth caretaker manager Lee Bradbury told BBC Radio Solent: "They are a good side and I thought we made them look decidedly average.
"It was a great goal for a fitting occasion - I had goosepimples all over when (Liam Feeney) finished that one.
"We defended really well - two clean sheets in two games so that gives the lads a confidence boost."
Brighton boss Gus Poyet told BBC Sussex: "I feel bad because we were a little bit too slow today. We thought sooner or later we'd have chances and we were just waiting. Sometimes you convince the players to be patient and maybe today we missed a bit of acceleration.
"You have to give credit to them because they planned the game and they played one way. We put them under pressure at the end. They did what they had to do and they beat us.
"The results today were quite scary. I don't know if the opposition don't want to catch us but they're doing everything possible not to."
