After conceding five last weekend, the visitors went behind when Lee Barnard smashed home Rickie Lambert's pass.
Jose Fonte headed in Ryan Dickson's cross, before Saints added a third, Dean Hammond finding Richard Chaplow, who volleyed in from eight yards.
Posh got a consolation through Aaron Mclean's run and tidy finish, but the rout was complete when Guly Do Prado headed in Lee Holmes' free-kick.
Nigel Adkins told BBC Radio Solent: "Four goals, it could have been a lot more, their goalkeeper made some excellent saves.
"I thought the backing from our supporters was magnificent.
"It takes a lot to get me off my seat...and I was jumping after a couple of them...their goalkeeper made lots of saves."
Peterborough United boss Gary Johnson told BBC Radio Cambridgeshire: "Southampton came out the traps well and all sections didn't perform today.
"We didn't defend properly, we didn't win the midfield battle and we didn't hold it up upfront.
"The best team won on the day and we've got a little bit of work to do."
