Southampton beat south coast neighbours Bournemouth in the sides' first league meeting for 50 years as Rickie Lambert scored twice from the penalty spot.
Bournemouth's Liam Feeney hit the post early on but when Rhoys Wiggins slipped and handled on the ground, Lambert smashed his penalty past Shwan Jalal.
After the break, Marvin Bartley tripped Morgan Schneiderlin and Lambert made no mistake with his second spot-kick.
Cherries defender Adam Smith was sent off for a second booking late on.
Bournemouth had reshuffled at half-time, bringing Shaun Cooper on for Feeney - and the utility man fired just over the bar as they tried desperately to get back into the game.
But Saints remained in command as Jalal tipped away a Lambert free-kick from the edge of the box as the Saints striker searched for his hat-trick.
The result was a repeat of Saints' 2-0 Carling Cup win at St Mary's in August, and the home side's long-awaited first home league win of the season.
It also meant the Cherries dropped out of second place as Brighton - who Eddie Howe's side visit next Saturday - remain top of League One.
Southampton boss Nigel Adkins told BBC Radio Solent: "It's a positive, we got our first victory at St Mary's, a victory against a local team who are flying high in the division and I thought we were comfortable.
"In the first 15 minutes ,the game should have been dead and buried with the amount of chances we had before our first penalty.
"For me, we've controlled the game. It's pleasing that we got a victory, a clean sheet, we move up the table and there's a lot of positives to take from it."
Bournemouth boss Eddie Howe told BBC Radio Solent: "I was disappointed with the performance. People will talk about the referee and I will probably say my bit. But we have look at ourselves first and foremost.
"We didn't play as well as we have done or as well as we can do.
"When you defend as well as we did and restrict their chances, to be defeated by two spot kicks which we felt were both debatable is disappointing."
