Ian Henderson struck a late winner for Colchester to leave Rochdale waiting for their first League One victory.
The U's had already hit the crossbar when Andrew Bond's shot deflected off Jason Kennedy to beat Josh Lillis and put them ahead.
Richard Dawson met Brian Barry-Murphy's corner to drive home Rochdale's equaliser from eight yards out.
But they were denied a point in the 84th minute when Henderson dinked the ball over Lillis.
Rochdale manager Keith Hill told BBC Radio Manchester: "I thought we should have at least got a point from the game, but I thought the way we played was good against a very good side.
"We've got to be a little bit more ruthless in pursuit of trying to score and there has got to be a balance of going in pursuit of winning games and not losing them.
"I'm relatively pleased with the performance especially with the ball but without the ball we can't be as naive as we have been today."
Colchester United manager John Ward told BBC Essex: "I have seen two very good football teams today, and I'm sure the people of Rochdale will enjoy watching their side.
" He (Ian Henderson) owed me that after he hit the bar in the first half, when he should have done better with that, so we'll call it quits!"
"Really pleased to at this stage to have two draws and a win, we have had to work really hard to take these points."
