Kevin Lisbie scored a stoppage-time winner to see off Leyton Orient and end Colchester's disappointing League One campaign with maximum points.
Lisbie struck from the penalty spot after Luke Ashworth handled Kem Izzet's goal-bound effort and was sent off.
Earlier, Kayode Odejayi dragged a shot wide for the hosts and Orient's Sean Thornton nodded over from close range.
Thornton cleared Phil Ifil's diving header off the line before Lisbie held his nerve from 12 yards.
Colchester United manager Aidy Boothroyd told BBC Essex:
"I'm glad we got the win for the fans. I think that is important.
"We will go home and have a drink tonight and we'll ask what more could we have done. we won, but we want to improve and that is what we will do next season."
Leyton Orient manager Russell Slade told BBC London 94.9:
"It wasn't the result I expected, to be honest I didn't think anybody was going to go on a win the game."
"I thought we were worth something today."
"The fans were magnificent today and the team appreciated that."
