James Dunne sealed Exeter's first win since January with a last-minute goal.
The midfielder headed in at the near post from Ryan Harley's corner to claim a much-needed three points for Exeter in their battle to avoid relegation.
Rovers came closest to scoring in the first half when Byron Anthony let fly from 35 yards and hit the bar.
Adam Stansfield missed two good chances for Exeter, the best of which a shot that went wide of goal from 10 yards, which he should have done better with.
Exeter City manager Paul Tisdale told BBC Radio Devon: "A fantastic evening for us and much deserved and I think it was one hell of a team performance.
"The crowd were magnificent today, they were positive from the first to the last, they were brilliant.
"We got it with a set-piece that we've been practicising so well done to everyone."
