Southampton sealed their first league win of the season courtesy of two penalties from Rickie Lambert.
The Liverpudlian opened the scoring on 18 minutes after being upended by Glovers defender Terrell Forbes.
Bottom-placed Saints remained nervy, but sealed the points when Lambert converted a second penalty after a Nathan Jones handball.
Defeat leaves Yeovil, without a win in nine games, still seeking a first away victory under manager Terry Skiverton.
Southampton manager Alan Pardew told BBC Radio Solent:
"It was a great win for us and probably will be the most important victory of the season
"It was good to get the second goal, it seemed a bit harsh on Yeovil but perhaps that's what we needed to lift the pressure
"We saw the game out well and the tactical decisions we made worked for us
Yeovil Town player-manager Terry Skiverton told BBC Somerset:
"I wouldn't mind if it was Man Utd away and people feel intimidated but when it's Southampton it calls into question what they do outside of football to handle the pressure.
"I thought my boys played really well, I think we played the better football and created some good chances in the first half but couldn't quite take them.
"I think leading up to the second goal we were on top again and then that happens and you can't quite prepare yourself for that in any team talk.
" We're missing one little leader in there to bring it together and I really don't think we're that far away from being a good side.
