Paul McLaren and Antony Kay scored first-half goals as Tranmere inflicted Carlisle's fourth defeat of the season. The home team wasted several chances before McLaren struck with a 20-yard finish following a corner from Jennison Myrie-Williams.
Kay then volleyed Tranmere's second from 18 yards before the break after Tranmere failed to clear a corner.
Carlisle's Chris Greenacre struck the post while McLaren cleared a Danny Livesey header off the line.
